Pepa Rus made her television debut in mid-2007, joining the secondary cast of the series Aída. She took on the role of Inmaculada "Macu" Colmenero, the young niece of the character Mauricio. This opportunity came as a surprise to Rus, who mentioned learning about it while on a bus trip to Seville to visit her family. To prepare for the role, she dedicated herself to watching all the episodes of 7 vidas and the initial seasons of Aída, indicating her commitment to understanding the show's context and her character's background. She described the characters in Aída as "flawed characters, like all of us," suggesting an appreciation for the show's realistic portrayal of individuals.

In 2010, Pepa Rus made her cinematic debut in the film La mula_{La Mula}^Spanish (The Mule), where she played the character Josefa "Pepi." In this film, she shared the screen with notable Spanish actors such as Mario Casas, María Valverde, and Secun de la Rosa. Her role in La mula was a supporting one, contributing to the ensemble cast of the production.

In 2015, she joined the cast of Gym Tony_{Gym Tony}^Spanish, where she portrayed Secundina "Secun" García.
