In 2009, he participated in the sprint at the Hovedlandsrennet in Beitostølen, where he earned a medal in the relay event, competing alongside Jarle Midthjell Gjørven, Runar Netland, and Johan Eirik Meland. Later the same year, he showcased his early dominance by winning two gold medals in the Men's 17 category at the Norwegian National Championships in roller ski shooting, triumphing in both the sprint and pursuit events held in Vik i Sogn. His international success began to materialize in 2010 and 2011, during which he secured multiple medals. By 2012, Bø had become a three-time junior world champion, a remarkable achievement that led to his inclusion in the Norwegian senior national team.

The 2018-2019 season marked Johannes Thingnes Bø's emergence as a truly dominant force in the Biathlon World Cup. He continued his extraordinary performance into the new year, achieving podium finishes in every race leading up to the events in Soldier Hollow.
At the 2019 Biathlon World Championships in Östersund, Sweden, Bø further solidified his World Cup dominance by winning the sprint event. He also added three gold medals in team events, showcasing his versatility and strength across different formats. During this season, Bø set a new record for the most individual victories in a single World Cup season, achieving 16 wins and surpassing the previous record of 14 held by Martin Fourcade.

Highlights of his season included significant victories in Hochfilzen, Le Grand-Bornand, and Nové Město. He also delivered a strong performance at the 2020 Biathlon World Championships in Antholz-Anterselva, Italy, where he secured six medals, including an individual gold in the mass start event.
The season concluded amidst the COVID-19 pandemic, which led to events in Kontiolahti being held without spectators and others being canceled. In a dramatic final race, Bø secured the overall World Cup title, narrowly defeating his retiring rival Martin Fourcade by a mere two points.
